Solifenacin Succinate – Medical Description

Solifenacin succinate is an oral antimuscarinic (anticholinergic) agent indicated for the management of overactive bladder (OAB). It works by selectively blocking muscarinic (M3) receptors in the urinary bladder, reducing involuntary bladder contractions, increasing bladder capacity, and decreasing episodes of urinary urgency, frequency, and urge urinary incontinence. Solifenacin is administered once daily and has demonstrated efficacy in improving urinary symptoms and quality of life in appropriately selected patients.

Medical Benefits

  • Reduces urinary urgency.
  • Decreases daytime urinary frequency.
  • Reduces urge urinary incontinence (urine leakage).
  • Increases functional bladder capacity.
  • Improves quality of life in patients with overactive bladder.
  • Convenient once-daily dosing with sustained symptom control.

Mechanism of Action

Solifenacin competitively inhibits M3 muscarinic receptors located on the detrusor muscle of the urinary bladder. This decreases involuntary bladder contractions during the filling phase, allowing greater urine storage and reducing symptoms of overactive bladder.

Adult Use

Indications:

  • Overactive bladder with symptoms of:
    • Urinary urgency
    • Increased urinary frequency
    • Urge urinary incontinence

Usual Adult Dose

  • 5 mg orally once daily.
  • May be increased to 10 mg once daily based on clinical response and tolerability.
  • Can be taken with or without food.
  • Tablets should be swallowed whole with water.

Pediatric Use

  • Routine use is not recommended for children with idiopathic overactive bladder unless prescribed by a pediatric urologist or specialist.
  • Some countries have approved pediatric formulations of solifenacin for neurogenic detrusor overactivity (NDO) in children, while standard 5 mg and 10 mg tablets are generally not approved for routine pediatric use.
  • Safety and dosing in children should always be determined by a specialist according to the approved indication and body weight.

Common Adverse Effects

  • Dry mouth
  • Constipation
  • Blurred vision
  • Dry eyes
  • Indigestion
  • Urinary retention (rare)
  • Dizziness or drowsiness in some patients

Contraindications

Do not use solifenacin in patients with:

  • Urinary retention
  • Gastric retention
  • Uncontrolled narrow-angle glaucoma
  • Known hypersensitivity to solifenacin or any component of the formulation.

Precautions

  • Use cautiously in patients with severe renal or hepatic impairment.
  • May impair the ability to drive or operate machinery due to blurred vision or drowsiness.
  • Use with caution in elderly patients because anticholinergic effects may be more pronounced.
  • Potential interactions may occur with strong CYP3A4 inhibitors (e.g., ketoconazole).

Storage

  • Store at 20–25°C (68–77°F).
  • Protect from excessive heat and moisture.
  • Keep out of the reach of children.
