Based on authoritative pharmaceutical and medical databases (including NIH, MDPI, and DrugBank), Mebeverine is a musculotropic antispasmodic primarily used for the symptomatic relief of Irritable Bowel Syndrome (IBS) and other gastrointestinal spasms. 

Mechanism of Action
Mebeverine works directly on the smooth muscles of the gastrointestinal tract to relieve spasms. 

  • Dual Action: It acts by blocking voltage-gated sodium channels and inhibiting intracellular calcium accumulation, which helps normalize bowel motility.
  • Targeted Relief: Unlike some other antispasmodics, it typically does not produce systemic "anticholinergic" side effects like dry mouth or blurred vision at standard doses. 

Common Dosing & Administration
The medication is most effective when taken before meals to manage post-prandial symptoms. 

  • Adult Standard Dose: Typically 135 mg taken three times daily.
  • Modified Release: 200 mg capsules are usually taken twice daily.
  • Timing: It should be taken approximately 20 minutes before meals and swallowed whole with water (do not chew). 

Safety and Side Effects
Mebeverine is generally well-tolerated, with a safety profile often comparable to a placebo in clinical trials. 

  • Frequent Side Effects: Indigestion, heartburn, dizziness, insomnia, and mild skin rashes or hives.
  • Serious Reactions: Rare hypersensitivity reactions, including angioedema (swelling of the face/neck) and anaphylaxis, have been reported.
  • Drug Tests: It can rarely cause a false positive for amphetamines or "ecstasy" in certain urine drug screenings. 

Key Precautions

  • Pregnancy & Breastfeeding: Not recommended during pregnancy as safety data is limited. While it passes into breast milk in small amounts, its use during breastfeeding is generally avoided by manufacturers.
  • Contraindications: Should not be used in cases of paralytic ileus (where the bowel is inactive) or by individuals with known allergies to any of its ingredients.
  • Pediatric Use: Generally not recommended for children under 18 years old, as its efficacy in this group has been found to be similar to placebo in some studies.
