Composition:- 

Each 5 g. (sachet) Contains:- 

- Active ingredients: 

- Piperazine Citrate ------------ 0.1408 g. ( Eq. To 0.128 g. Piperazine Hydrate )

 - Colchicine Crystals ---------- 0.3 mg. 

 - Atropine Sulphate ------------ 0.128 mg.

- Inactive Ingredients: Sod.Bicarbonate, Citric acid anhy., Tartaric acid, P.V. P, Lemon oil, Sucrose.

Pharmaceutical Form:- 

Effervescent Granules.

Pharmacological Action:-  

• An acute attack of gout apparently occurs as a result of an inflammatory reaction to crystals of monosodium urate that are deposited in the joint tissue.

• Piperazine known for many years to be an active drug in the treatment of gout and urinary lithiasis. It is claimed to increase the solubility of urate in the urine. It is relatively non- toxic.

• Colchicine gives rapid relief from pain in acute attacks of gout and the drug is specific for this condition to the extent that it can be used as a diagnostic agent in gout and gouty arthritis. Colchicine possess anti-inflammatory properties. Although it is highly effective in treating acute gouty arthritis, it is not effective for other types of pain. It is not an analgesic and does not affect uric acid clearance. Instead, colchicine binds to proteins in microtubules of neutrophils. This binding, in turn, inhibits the migration of neutrophils into the area of inflammation, thereby interfering with the inflammatory response to urate crystal deposition. 

• Atropine sulphate is added to relieve the spasm of the urinary tract.

Pharmacokinetics:- 

- Urosolvine contains a typical uricosuric agent that promotes the urinary excretion of uric acid and thereby lower serum uric acid concentration. 

 - Sodium citrate is an alkalinising agent since the citrate is converted into bicarbonate; alkaline urine is definitely more suitable for the excretion of urates in a more soluble form. 

Indications:-  

- Acute and chronic gout.

 - Gouty arthritis. 

 - For prophylaxis against urinary lithiasis ( urate stones). 

Dosage and Administration:- 

Dissolve the contents of one packet in a half glass of water before meals 3 times daily or as prescribed by the physician. 

Contraindications:-

- Known hypersensitivity to any of the components of the drug. 

 - Blood dyscrasias.

 - Acute dehydration, anuria or hyperkalemia. 

Side Effects:- 

Occasionally nausea, vomiting, abdominal pain and diarrhea may occur.

Drug Interaction:- 

None.

Pregnancy and Lactation:- 

- Contraindicated in pregnant women.

 - It is unknown if drug appears in breast milk.

Precautions and Warning:- 

Used with caution: in patients with serious GIT, renal, hepatic, and cardiovascular disorders, in children, geriatrics, debilitated patients and nursing women.

Package and Storage:- 

- Box of 12 SachetsX 5 g. and leaflet.

 - Store below 25 ºC in a dry place.

Instruction to Patients:- 

- Reduce dosage if vomiting or diarrhea occurs.

 - Keep out of reach of children.
